IMP is good, but if you're not running your own e-mail server or if you can't allocate a POP/IMAP account to each user, you might like to have a look at VAMP at http://undead.thegraveyard.org/vamp.html
You have all mail sent to anyaddress@yourdomain delivered to a single POP account, and VAMP then examines the headers to determine which messages are for which of your "virtual users". I've set up a (highly customised) version for a new site I'm doing. I've not thoroughly tested its reliability yet, but it's a good stopgap solution for us, until we get big enough to outsource that side of things.
Because it uses a mySql database, we've been able to completely integrate account creation into the registration pages for our site quite easily.
The one thing I would say is that VAMP is probably only suitable for a relatively small number of users (maybe under one hundred?)
